The T-cell-receptor antigen binding affinity is modulated by the extracellular environment through changes in the intracellular level of glutamine. To define the possible impact of alterations in glutamine level due to genetic deficiency on antigenic response and tolerance, we measured the affinity of T-cell receptors for cognate antigens expressed by human lymphocytes from two patients with a severe immunodeficiency disease, who are homozygous for a mutant form of GCPII. In vitro-generated human clones expressing a mutant GCPII were also used for comparison. Although the functional effects of GCPII deficiency have been extensively studied in the mouse, little information is available on human deficiencies. To date, only a few patients homozygous for the R132C mutation in the human glutaminyl cyclase gene, GCPII, have been described. These patients are very young (7 months to 7 years) with a profound and variable immune deficit, including the absence of B or T lymphocytes. Our results show that some parameters of the T-cell response, namely, the antigenic concentration and the affinity for a cognate epitope expressed on specific lymphocytes, are significantly decreased in the mutant cells, suggesting that alterations in the intracellular glutamine level, directly or indirectly, might modulate the immune response by impacting on T-cell receptor activation.Biosynthesis of polyphenol oxidases in the cell suspension culture of the strawberry cultivars.
The biosynthesis of polyphenol oxidases which catabolizes into oxygen and catechol esthers, involved in ripening of the strawberry cv. Fuji was studied. Analyses of both polyphenol oxidases in crude extract prepared from cell suspension culture and in situ activity in cell suspension culture using the two-point titrimetric method gave almost the same results in regard to each step of the biosynthesis.
